Rate Measurement of Single Budding Yeast

The development of new technologies to manipulate and analyze single cells is a crucial goal of modern biotechnology. Microfluidic lab-on-a-chip technology provides unique opportunities to interrogate a small population of or even single cells. In this talk, I will present measurements of the growth rate of single budding yeasts with two microfluidic devices and the development of one microfluidic device that can sort and detect yeast cells of different sizes simultaneously.
The first device that enables the dynamic measurement of the volume growth rate of a single cell is a MOSFET-based microfluidic Coulter-type sensor. By moving a budding yeast cell back and forth through the sensing channel and measuring the induced resistance modulation from the translocation of the budding yeast, the volume growth rate of the single budding yeast cell can be measured over its whole cell cycle. The second microfluidic device consists of two identical microfluidic channels: one sensing channel and one reference channel. This sensing scheme helps to remove any fluctuation and draft in the electrical and fluidic circuit, which enables the measurement of cell growth rate in the sensing channel in real time. The microfluidic device that can separate yeast cell colony according to the cell size is realized through dielectrophoresis in the non-uniform electric field created by a sharp insulating hurdle in the microfluidic circuit. This microfluidic cell separation circuit is integrated with a MOSFET-based resistive pulse sensing circuit at the downstream to realize simultaneous on-chip separation and sizing of the yeast cells.

Ms. Jiashu Sun is currently a Ph.D. candidate in the Department of Mechanical Engineering at Vanderbilt University. Her research interest is in microfluidics for biological and biomedical applications.
