Recognition and modification of small silencing RNAs: structures and mechanisms

RNA silencing is a conserved regulatory mechanism in fungi, plants and animals to regulate gene expression and defense against viruses and transgenes. Small silencing RNAs of ~20 to 30 nucleotides (nt) and their associated effector proteins, members of the Argonaute family proteins, are the central components in RNA silencing. Although a rapidly growing number of small silencing RNAs have emerged, most of them belong to three classes based on their distinct structures and biogenesis: small interfering RNAs (siRNAs), microRNAs (miRNAs) and Piwi-interacting RNAs (piRNAs). A subset of small RNAs, such as miRNAs and siRNAs in plants, piRNAs in animals and siRNAs in Drosophila, require an additional crucial step for their maturation, 2’-O-methylation on the 3’ terminal nucleotide. A conserved S-adenosyl-L-methionine (AdoMet)-dependent RNA methyltransferase, HUA ENHANCER 1 (HEN1), and its homologs are responsible for this specific modification. Based on structural and biochemical studies in my lab, I will present the molecular basis for the recognition and molecular mechanism for the 3’-end methylation of small silencing RNAs.
